    Abstract The longevity and high activity of the cork cambium (or phellogen) from Quercus suber L. (cork oak) are the cornerstones for the sustainable exploitation of a unique raw material. Cork oak is a symbolic model to study cork development and cell wall suberization, yet most genetic and molecular studies on these topics have targeted other model plants. In this study, we explored the potential of taproots as a model system to study phellem development and suberization in cork oak, thereby avoiding the time constraints imposed when studying whole plants. In roots, suberin deposition is found in mature endodermis cells during primary development and in phellem cells during secondary development. By investigating the spatiotemporal characteristics of both endodermis and phellem suberization in young seedling taproots, we demonstrated that secondary growth and phellogen activity are initiated very early in cork oak taproots (approx. 8 days after sowing). We further compared the transcriptomic profile of root segments undergoing primary (PD) and secondary development (SD) and identified multiple candidate genes with predicted roles in cell wall modifications, mainly lignification and suberization, in addition to several regulatory genes, particularly transcription factor- and hormone-related genes. Our results indicate that the molecular regulation of suberization and secondary development in cork oak roots is relatively conserved with other species. The provided morphological characterization creates new opportunities to allow a faster assessment of phellogen activity (as compared with studies using stem tissues) and to tackle fundamental questions regarding its regulation.

NOV1 is a dioxygenase that catalyzes the one-step, coenzyme-free oxidation of isoeugenol into vanillin and holds enormous biotechnological potential for the complete valorization of lignin as a sustainable starting material for biobased chemicals, polymers, and materials. This study integrates computational, kinetic, structural, and biophysical approaches to characterize a new NOV1 variant featuring improved activity and stability compared to those of the wild type. The S283F replacement results in a 2-fold increased turnover rate (kcat) for isoeugenol and a 4-fold higher catalytic efficiency (kcat/Km) for molecular oxygen compared to those of the wild type. Furthermore, the variant exhibits a half-life that is 20-fold higher than that of the wild type, which most likely relates to the enhanced stabilization of the iron cofactor in the active site. Molecular dynamics supports this view, revealing that the S283F replacement decreases the optimal pKa and favors conformations of the iron-coordinating histidines compatible with an increased level of binding to iron. Importantly, whole cells containing the S283F variant catalyze the conversion of ≤100 mM isoeugenol to vanillin, yielding >99% molar conversion yields within 24 h. This integrative strategy provided a new enzyme for biotechnological applications and mechanistic insights that will facilitate the future design of robust and efficient biocatalysts. publishersversion published

Neste artigo procuro fazer algumas dessas interpretações sobre o intercâmbio entre o pictórico e o literário que ocorre na obra de Almada Negreiros, com ênfase precisamente no intercâmbio, isto é, não notando meramente a criação de imagens visuais literárias — o que tem sido o foco dos estudos literários — mas entendendo que a escrita de Almada deriva de uma concepção de vanguarda e de arte assente no primado da visão e que a sua prática e teoria pictóricas determinam a sua escrita. Por outro lado, abordarei também a revisitação que faz em pintura da sua obra literária. Mais do que um diálogo entre duas artes, o pictórico e o literário em Almada estão profundamente interligados e a obra daqui resultante constitui-se um exemplo da inventividade que resultou da ruptura de fronteiras entre as artes operada pelas vanguardas do início do século. Many authors have recognized the visual character of the writings by the Portuguese artist Almada Negreiros (1893-1970), but the multiple variations and interpretations that can be driven from that are yet to be explored. In this article I intend to make some of those interpretations about the interchange between the pictorial and the literary that occur in his work, emphasizing precisely the interchange, that is, not noticing merely how he creates strong literary images — which has been the focus of the literary studies — but understanding that Almada’s writing thrives from a notion of avant-garde based on the primacy of vision and that his practice and theory about the pictorial are determinant for his writing. On another hand, I will also consider how his painting often revisits his literary work. More than a dialogue between two art forms, the pictorial and the literary in Almada Negreiros are profoundly interconnected and constitute an example of the inventiveness that resulted from the rupture of the frontiers between the arts operated by the avant-garde from the early twentieth century. publishersversion published

    Long-term cognitive dysfunction, or “chemobrain”, has been observed in cancer patients treated with chemotherapy. Mitoxantrone (MTX) is a topoisomerase II inhibitor that binds and intercalates with DNA, being used in the treatment of several cancers and multiple sclerosis. Although MTX can induce chemobrain, its neurotoxic mechanisms are poorly studied. This work aimed to identify the adverse outcome pathways (AOPs) activated in the brain upon the use of a clinically relevant cumulative dose of MTX. Three-month-old male CD-1 mice were given a biweekly intraperitoneal administration of MTX over the course of three weeks until reaching a total cumulative dose of 6 mg/kg. Controls were given sterile saline in the same schedule. Two weeks after the last administration, the mice were euthanized and their brains removed. The left brain hemisphere was used for targeted profiling of the metabolism of glutathione and the right hemisphere for an untargeted metabolomics approach. The obtained results revealed that MTX treatment reduced the availability of cysteine (Cys), cysteinylglycine (CysGly), and reduced glutathione (GSH) suggesting that MTX disrupts glutathione metabolism. The untargeted approach revealed metabolic circuits of phosphatidylethanolamine, catecholamines, unsaturated fatty acids biosynthesis, and glycerolipids as relevant players in AOPs of MTX in our in vivo model. As far as we know, our study was the first to perform such a broad profiling study on pathways that could put patients given MTX at risk of cognitive deficits.

    Abstract Ecological models predict that the effects of mammalian herbivore exclusion on plant diversity depend on resource availability and plant exposure to ungulate grazing over evolutionary time. Using an experiment replicated in 57 grasslands on six continents, with contrasting evolutionary history of grazing, we tested how resources (mean annual precipitation and soil nutrients) determine herbivore exclusion effects on plant diversity, richness and evenness. Here we show that at sites with a long history of ungulate grazing, herbivore exclusion reduced plant diversity by reducing both richness and evenness and the responses of richness and diversity to herbivore exclusion decreased with mean annual precipitation. At sites with a short history of grazing, the effects of herbivore exclusion were not related to precipitation but differed for native and exotic plant richness. Thus, plant species’ evolutionary history of grazing continues to shape the response of the world’s grasslands to changing mammalian herbivory.

While pyrethroids are recommended only to reduce/limit transmission, they are widely implemented to reduce biting nuisance and to control agricultural pests, increasing the risk of insurgence of resistance mechanisms. Worryingly, pyrethroid resistance (with mortality < 70%) was recently reported in Ae. albopictus populations from Italy and Spain and associated with the V1016G point mutation in the voltage-sensitive sodium channel gene conferring knockdown resistance (kdr).     Bergstrom, Blume and Varian (1986) provides an elegant game-theoretic model of an economy with one private good and one public good. Strategies of players consist of voluntary contributions of the private good to public good production. Without relying on first order conditions, the authors demonstrate existence of Nash equilibrium and an extension of Warr's neutrality result -- any redistribution of endowment that left the set of contributors unchanged would induce a new equilibrium with the same total public good provision. The assumption of one-private good greatly facilities the results. We provide analogues of the Bergstrom, Blume and Varian results in a model allowing multiple private and public goods. In addition, we relate the strategic market game equilibrium to the private provision of equilibrium of Villanaci and Zenginobuz (2005), which provides a counter-part to the Walrasian equilibrium for a public goods economy. Our techniques follow those of Dubey and Geanakoplos (2003), which itself grows out of the seminal work of Shapley and Shubik (1977). Our approach also incorporates, into the strategic market game literature, economies with production, not previously treated and, as a by-product, establishes a new existence of private-provision equilibrium.

    Keeping replicated data strongly consistent is convenient when communication is fast and available. In internet-scale distributed systems the reality of high communication latencies and likelihood of partitions, leads developers to adopt more relaxed consistency models, such as eventual consistency. Conflict-free Replicated Data Types, bring structure to the design of eventually consistent data management solutions, by precisely describing the behaviour under concurrent updates and guarantying a path to reconciliation. This paper offers a survey of the mathematical structures that support state based multi-master replication with reconciliation, and shows how state structures and state transformations can be composed to provide data types that are now used in practice in many geo-replicated systems.     Due to the inappropriate use of antibiotics described in both human and veterinary medicine, there is emerging evidence of antimicrobial-resistant organisms isolated from humans and pets, forming a multifaceted problem. Although the true magnitude of antimicrobial resistance in pets and other animals, as well as humans, are not fully known; pets, in particular dogs and cats, can contribute to the spread of antimicrobial resistance due to close contact with humans and their status as a family member in urban households. This review summarizes and highlights the current data concerning the antibiotic use on pets, and the European distribution of the increasing prevalence of multiresistant bacterial pathogens, such as enterococci and methicillin-resistant staphylococci on pets, as well as its implications for public health.

    UID/ANT/04038/2019 IF/00127/2014 Ongoing multidisciplinary studies of skeletonized human remains from the Middle Neolithic Bom Santo Cave (Lisbon, Portugal) is indicating a very heterogeneous population at various levels (diets, mobility and genetics). The current interpretation suggests that its socioeconomic and funerary territories encompassed the lower Tagus, its tributaries and the granitic sectors of the Mora–Pavia area in the Alentejo. Archaeothanatological analyses indicated mutually exclusive funerary practices: secondary depositions at Room A and primary and secondary depositions at Room B. Polished stone tools are evenly distributed in both rooms, while ornaments, pottery, flint blades and sheep/goat phalanges are almost restricted to Room A. Such distribution patterns reflects the coexistence of distinct funerary practices in which Room A is part of a much complex behaviour that included primary depositions, exhumation, transportation and re-deposition of human bone remains between different sectors of the cave and/or cemeteries (caves, dolmens) of the above-described territory. Thus, a more dynamic (in its rituals) and wider (in its geography) set of funerary practices than usually perceived — in which the intentional segmentation of human skeletons is attested — seems to have taken place at the onset of megalithism in central-southern Portugal. publishersversion published
