    The nitrite adducts of globins can potentially bind via O- or N- linkage to the heme iron. We have used EPR (electron paramagnetic resonance) and DFT (density functional theory) to explore these binding modes to myoglobin and hemoglobin. We demonstrate that the nitrite adducts of both globins have detectable EPR signals; we provide an explanation for the difficulty in detecting these EPR features, based on uniaxial state considerations. The EPR and DFT data show that both nitrite linkage isomers can be present at the same time and that the two isomers are readily interconvertible in solution. The millisecond-scale process of nitrite reduction by Hb is investigated in search of the elusive Fe(II)-nitrite adduct. Highlights • A DFT-derived barrier for nitrite linkage isomerism on heme center is reported. • EPR spectra of nitrite adducts show evidence for linkage isomerism. • The electronic structure of Fe(III)-nitrite heme is conformation-dependent. • Certain conformations are inducive to EPR silence. • Fe(II)-nitrite is undetectable on stopped-flow time scales.     AbstractPolymerase Chain Reaction (PCR) is the principal method of amplifying target DNA regions and, as such, is of great importance when performing microbial diversity studies. An unfortunate side effect of PCR is the formation of unwanted byproducts such as chimeras. The main goal of the work covered in this article is the development of an algorithm that simulates realistic chimeras for use in the evaluation of chimera detection software and for investigations into the accuracy of community structure analyses. Experimental data has helped to identify factors which may cause the formation of chimeras and has provided evidence of how influential these factors can be. This article makes use of some of this evidence in order to build a model with which to simulate the PCR process. This model helps to better explain the formation of chimeras and is therefore able to provide aid to future studies that intend to use PCR.

    The celebration of individual agency, which is closely connected to the promotion of the knowledge economy, competition and performance, is obvious in contemporary European education and youth policy, at national and supranational levels. Schools and teachers are thus expected to be crucial agents in fostering the development of flexible, responsible, self-governing and entrepreneurial citizens by adopting personalised pedagogies and continuous and close evaluation (Hartley, 2009). For example, the project Education Governance, Social Integration and Exclusion in Europe (EGSIE), which included studies from several European countries, revealed considerable consensus among teachers, principals and decision-makers on what constitutes a good student, a good teacher or a good head teacher, clearly reflecting such ideas about agency (Lindblad et al, 2002; Lindblad & Popkewitz, 2004). Consequently, structural and institutional power relations (Arnesen & Lundahl, forthcoming 2010) are downplayed and expressions of individual agency in terms of collective action and resistance are rendered more or less invisible. Furthermore, the emphasis on learning combined with high achievement and control may lead to more children and young people being marginalised and left with less room for agency, resulting in teachers’ attempts to promote socio-emotional development and fostering democratic citizenship being toned down. Such findings, however, make it no less essential to map and analyse new forms of influence and action, new freedoms and controls, individual as well as collective, resulting from or being in conflict with emerging power configurations This special issue highlights the subject of agency in Nordic educational institutions during the early 2000s, with respect to acting on, negotiating, opposing, transgressing and resisting, as well as examining the options for participation by various actors. It is suggested that altered relationships and increased tensions between individual ‘freedom’ (cf. Rose, 1999) and institutional control, as well as conflicting policies and values, bring about particular power dynamics, through which different actors within institutions may gain more or less influence. We also inquire into disruptions or actions that go against the grain. Whilst studies focusing on resistance often regard teachers and students as conflicting groups (see McFadden, 1995), our aim is also to search for alliances that might involve various constellations of students, teachers, and other actors. We look for contradictions in critical manifestations, since the position of being critical and in opposition is more open for some actors, e.g. middle-class boys and male teachers with permanent posts, than

    Dark littoral submarine caves can act as enclaves of the deep aphotic zone in shallow coastal areas, and their survey has revealed the existence of a very particular fauna of specialized and poorly known organisms among which crustaceans are particularly well represented. In these particular habitats, the use of conventional sampling techniques, such as hand nets, is often not recommended since they disturb bottom sediments causing hazardous situations to scientific divers. The use of baited traps, while technically possible, is not. always practical is such remote habitats. The present work describes a simple and inexpensive manual device that can be operated by divers ill submarine caves and other cryptic habitats to recurrently catch small motile organisms such as mysid crustaceans, caridean shrimps, or even gobiid fishes. This small suction bottle derived and improved from the original "Sket bottle" design considerably reduces the risks of disturbing the cave's bottom sediment and can be easily operated using a single hand. The described sampling device can also be easily used outside caves, in a variety of particular habitats, e.g., rubble filled bottoms, branching coral reefs, cracks, and small holes on rocky surfaces, in which small motile organisms usually escape from traditional sampling gears, e.g., fishnets and traps, or simply go unnoticed by researchers during sampling. info:eu-repo/semantics/publishedVersion

    Purpose: Cetuximab is an EGFR-targeted therapy approved for the treatment of RAS wild-type (WT) metastatic colorectal cancer (mCRC). However, about 60% of these patients show innate resistance to cetuximab. To increase cetuximab efficacy, it is crucial to successfully identify responder patients, as well as to develop new therapeutic approaches to overcome cetuximab resistance. Experimental design: We evaluated the value of EGFR effector phospholipase C gamma 1 (PLCγ1) in predicting cetuximab responses, by analyzing progression-free survival (PFS) of a multicentric retrospective cohort of 94 treated patients with mCRC (log-rank test and Cox regression model). Furthermore, we used in vitro and zebrafish xenotransplant models to identify and target the mechanism behind PLCγ1-mediated resistance to cetuximab. Results: In this study, levels of PLCγ1 were found increased in RAS WT tumors and were able to predict cetuximab responses in clinical samples and in vitro and in vivo models. Mechanistically, PLCγ1 expression was found to bypass cetuximab-dependent EGFR inhibition by activating ERK and AKT pathways. This novel resistance mechanism involves a noncatalytic role of PLCγ1 SH2 tandem domains in the propagation of downstream signaling via SH2-containing protein tyrosine phosphatase 2 (SHP2). Accordingly, SHP2 inhibition sensitizes PLCγ1-resistant cells to cetuximab. Conclusions: Our discoveries reveal the potential of PLCγ1 as a predictive biomarker for cetuximab responses and suggest an alternative therapeutic approach to circumvent PLCγ1-mediated resistance to cetuximab in patients with RAS WT mCRC. In this way, this work contributes to the development of novel strategies in the medical management and treatment of patients with mCRC. M.     Several lines of evidence suggest that the warming climate plays a vital role in driving certain types of extreme weather. The impact of this warming and extreme weather on forest carbon assimilation capacity is poorly known. Filling this knowledge gap is critical towards understanding changing the amount of carbon that forests can hold. Here, we used a perfect-deficit approach to identify forest canopy photosynthetic capacity (CPC) deficits and analyze how they correlate to climate extremes, based on data measured by the eddy covariance method at 26 forest sites integrating 146 site-years. We found that droughts severely affect the carbon assimilation capacities of evergreen broadleaf forest and deciduous broadleaf forest. In addition, the carbon assimilation capacities of Mediterranean forests are highly sensitive to climate extremes, while marine forest climates tend to be insensitive to climate extremes. Our estimates suggest an average global reduction of forest canopy photosynthetic capacity of 6.3petagramsof carbon per growing season over 2001-2010, with evergreen broadleaf forests contributing 51.7% of the total reduction.     We briefly review how microbial biotechnology can contribute to improve activities aiming to restore degraded drylands and to combat their desertification, which are an integral part of the Sustainable Development Goal 15 of the 2030 Agenda. Microbial biotechnology offers notable promise to improve restoration actions based on the use of biocrust-forming engineered cyanobacteria, which play key roles in maintaining ecosystem structure and functioning in drylands worldwide. Advances in our understanding of microbiome associated to biocrusts and of the signalling involved in the communication among their constituents can also potentially enhance the outcome of restoration activities in drylands.     Abstract. The Greenland ice sheet is one of the largest contributors to global meansea-level rise today and is expected to continue to lose mass as the Arcticcontinues to warm. The two predominant mass loss mechanisms are increasedsurface meltwater run-off and mass loss associated with the retreat ofmarine-terminating outlet glaciers. In this paper we use a large ensemble ofGreenland ice sheet models forced by output from a representative subset ofthe Coupled Model Intercomparison Project (CMIP5) global climate models to project ice sheet changes and sea-level risecontributions over the 21st century. The simulations are part of theIce Sheet Model Intercomparison Project for CMIP6 (ISMIP6). We estimate thesea-level contribution together with uncertainties due to future climateforcing, ice sheet model formulations and ocean forcing for the twogreenhouse gas concentration scenarios RCP8.5 and RCP2.6. The resultsindicate that the Greenland ice sheet will continue to lose mass in bothscenarios until 2100, with contributions of 90±50 and 32±17 mm to sea-level rise for RCP8.5 and RCP2.6, respectively. The largestmass loss is expected from the south-west of Greenland, which is governed bysurface mass balance changes, continuing what is already observed today.Because the contributions are calculated against an unforced controlexperiment, these numbers do not include any committed mass loss, i.e. massloss that would occur over the coming century if the climate forcingremained constant. Under RCP8.5 forcing, ice sheet model uncertaintyexplains an ensemble spread of 40 mm, while climate model uncertainty andocean forcing uncertainty account for a spread of 36 and 19 mm,respectively.     The widespread use of sensorial technologies has created new opportunities for enterprises, as these new sources of data assist in increasing the context-awareness in which enterprises operate and enable them to better anticipate and adapt to changes in the business environment. This leads to better decision-making and greater profitability, as well as to reduced operating risks. However, if enterprises are to get any benefit from these new data sources, there is a need for tools, which will enable them to deal with the complexity of the multitude of data sources, both from physical and virtual objects, as well as a means to extract relevant and correct information and knowledge from it. Having access to data is not enough. The real value to enterprises comes from being able to process it, interpret it and being able to make accurate forecasts upon which they can base their business decisions. This paper presents the blockchain technology which is intended to support the development of sensing enterprise systems for intelligent knowledge management. Since the creation of the internet, blockchain is the most important technology created that is in constant development and has still much more to develop. In a simple way, blockchain is a computational technology for register of operations, decentralized, free-access, transparent, global, continuous. It is a public database that is accessible for everyone and is much more secure and reliable than other forms currently known to perform similar operations. Thus, it is intended to demonstrate the importance of this recent technology in business processes and the future trends of its use in sensing enterprise business processes. The proposed framework intends to demonstrate and serve as the foundation for new business models supported by the new capabilities provided by sensorial technologies in the support of enterprise applications.

    AbstractApplications of LC-MS/MS for the biochemical diagnosis of catecholamine-producing tumors have followed from parallel emergence of this technology with changes in emphasis of biochemical testing, in particular requirements that testing include measurements of plasma or urinary fractionated metanephrines. Since the turn of the century, when LC-MS/MS was first introduced into the routine laboratory, there have been numerous advances in analytical methodology that offer opportunities for breaking away from out-moded analytical procedures to methods that provide enhanced diagnostic information with dramatically improved analytical sensitivity, accuracy and precision as well as rapid sample throughput. LC-MS/MS also offers high analytical specificity, but is not infallible. Multiple reaction monitoring does not always guarantee selectivity and attention to sample purification and chromatographic separation remains important. Although other analytical methods persist, today LC-MS/MS represents the method of choice for high-throughput, low-cost, precise and accurate measurements of the catecholamine metabolites now routinely used for diagnosis of catecholamine-producing tumors.

    Summary Background Bariatric surgery is associated with remission of diabetes and prevention of diabetic complications in patients with obesity and type 2 diabetes. Long-term effects of bariatric surgery on microvascular complications in patients with prediabetes are unknown. The aim of this study was to examine the effects of bariatric surgery on incidence of microvascular complications in patients with obesity stratified by baseline glycaemic status. Methods Patients were recruited to the Swedish Obese Subjects (SOS) study between Sept 1, 1987, and Jan 31, 2001. Inclusion criteria were age 37–60 years and BMI of 34 kg/m 2 or greater in men and 38 kg/m 2 or greater in women. Exclusion criteria were identical in surgery and control groups and designed to exclude patients not suitable for surgery. The surgery group (n=2010) underwent gastric bypass (265 [13%]), gastric banding (376 [19%]), or vertical-banded gastroplasty (1369 [68%]). Participants in the control group (n=2037) received usual care. Bodyweight was measured and questionnaires were completed at baseline and at 0·5 years, 1 year, 2 years, 3 years, 4 years, 6 years, 8 years, 10 years, 15 years, and 20 years. Biochemical variables were measured at baseline and at 2 years, 10 years, and 15 years. We categorised participants into subgroups on the basis of baseline glycaemic status (normal [fasting blood glucose concentration Findings 4032 of the 4047 participants in the SOS study were included in this analysis. We excluded four patients with suspected type 1 diabetes, and 11 patients with unknown glycaemic status at baseline. At baseline, 2838 patients had normal blood glucose, 591 had prediabetes, 246 had screen-detected diabetes, and 357 had established diabetes. Median follow-up was 19 years (IQR 16–21). We identified 374 incident cases of microvascular disease in the control group and 224 in the surgery group (hazard ratio [HR] 0·56, 95% CI 0·48–0·66; p Interpretation Bariatric surgery was associated with reduced risk of microvascular complications in all subgroups, but the greatest relative risk reduction was observed in patients with prediabetes at baseline. Our results suggest that prediabetes should be treated aggressively to prevent future microvascular events, and effective non-surgical treatments need to be developed for this purpose.
